Abstract

ABSTRAK Dislipidemia merupakan faktor risiko utama penyakit kardiovaskular yang ditandai oleh peningkatan kadar kolesterol total, low density lipoprotein (LDL), trigliserida, serta penurunan high density lipoprotein (HDL). Daun kelor (Moringa oleifera) mengandung senyawa bioaktif yang berpotensi memperbaiki profil lipid darah. Penelitian ini bertujuan menganalisis pengaruh pemberian ekstrak daun kelor terhadap kadar LDL dan trigliserida pada tikus model dislipidemia. Penelitian ini merupakan penelitian eksperimental dengan desain post-test control group. Tikus putih jantan galur Wistar diinduksi dislipidemia menggunakan HFD selama 28 hari, kemudian tikus dibagi ke dalam enam kelompok, yaitu kelompok kontrol normal (KN) yang diberi pakan standar tanpa perlakuan, kelompok kontrol negatif (K-) yang diberi HFD tanpa perlakuan, kelompok kontrol positif (K+) yang diberi HFD + simvastatin, serta tiga kelompok perlakuan yang masing-masing diberikan ekstrak daun kelor dengan dosis 250 mg/kgBB (P1), 500 mg/kgBB (P2), dan 750 mg/kgBB (P3) selama 14 hari. Pemberian HFD selama 28 hari secara signifikan meningkatkan kadar kolesterol total tikus. Analisis data menggunakan uji Welch Anova dan KruskalWallis dilanjutkan post hoc. Hasil uji Welch Anova menunjukkan terdapat perbedaan bermakna kadar LDL antar kelompok (p = 0,000). Uji lanjut Games– Howell pada kadar LDL menunjukkan bahwa kelompok kontrol negatif tidak berbeda bermakna dengan kelompok perlakuan dosis 250 mg/kgBB (p = 0,110), tetapi berbeda bermakna dengan kelompok dosis 500 mg/kgBB (p = 0,003) dan 750 mg/kgBB (p = 0,000). Hasil Uji Kruskal-Wallis menunjukkan terdapat perbedaan bermakna kadar trigliserida antar kelompok (p = 0,000). Uji lanjut Mann-Whitney pada kadar trigliserida menunjukkan bahwa kelompok kontrol negatif tidak berbeda bermakna dengan kelompok perlakuan dosis 250 mg/kgBB (p = 0,262), tetapi berbeda bermakna dengan kelompok dosis 500 mg/kgBB (p = 0,025) dan 750 mg/kgBB (p = 0,016). Penelitian ini menyimpulkan bahwa ekstrak daun kelor berpotensi sebagai agen hipolipidemik alami, dengan dosis 750 mg/kgBB merupakan dosis yang paling efektif dalam menurunkan kadar LDL dan trigliserida pada tikus model dislipidemia. Kata kunci: dislipidemia, daun kelor (moringa oleifera), LDL, trigliserida. ii ABSTRACT Dyslipidemia is a major risk factor for cardiovascular disease characterized by increased levels of total cholesterol, low-density lipoprotein (LDL), triglycerides, and decreased levels of high-density lipoprotein (HDL). Moringa oleifera leaves contain bioactive compounds that have the potential to improve blood lipid profiles. This study aims to analyze the effect of Moringa leaf extract on LDL and triglyceride levels in dyslipidemia model rats. This experimental study used post-test control group design. Male white Wistar rats were induced with dyslipidemia using HFD for 28 days, then the rats were divided into six groups, namely the normal control group (KN) which was given standard feed without treatment, the negative control group (K-) which was given HFD without treatment, the positive control group (K+) which was given HFD + simvastatin, and three treatment groups each given Moringa leaf extract at a dose of 250 mg/kgBW (P1), 500 mg/kgBW (P2), and 750 mg/kgBW (P3) for 14 days. Administering a HFD for 28 days significantly increased total cholesterol levels in mice. Data analysis using Welch Anova and Kruskal-Wallis tests followed by post hoc. The Welch Anova test showed a significant difference in LDL levels between groups (p = 0.000). A Games–Howell follow-up test on LDL levels showed that the negative control group was not significantly different from the 250 mg/kgBW dose treatment group (p = 0.110) but was significantly different from the 500 mg/kgBW (p = 0.003) and 750 mg/kgBW (p = 0.000) dose groups. The Kruskal-Wallis test showed a significant difference in triglyceride levels between groups (p = 0.000). Further Mann-Whitney test on triglyceride levels showed that the negative control group was not significantly different from the 250 mg/kgBW dose treatment group (p = 0.262) but was significantly different from the 500 mg/kgBW dose group (p = 0.025) and 750 mg/kgBW (p = 0.016). In conclusion, Moringa leaf extract has the potential as a natural hypolipidemic agent, with a dose of 750 mg/kgBW being the most effective dose in reducing LDL and triglyceride levels in dyslipidemia model rats.
