Abstract

ABSTRACT Pamelo (Citrus maxima (Burm.) Merr) is a fruit plant that has long been known in Indonesia. This orange has a distinctive character like its large fruit, has a fresh taste and long shelf life for up to four months. Propagation of pamelo citrus plants (Citrus maxima (Burm.) Merr) can not be done conventionally because pamelo seeds are difficult to germinate therefore must be done culture tissue. Plant tissue culture is one of the methods of propagation that can produce a uniform new plant in a relative shorter time while maintaining the sustainability of the parent plant. This research was conducted at the Tissue Culture Laboratory, Faculty of Agriculture, Malikussaleh University. This research was conducted from August to November 2020. The research method used is a factorial Randomized Block Design (RBD) that consist of two factors, concentration of coconut water (0%. 20%, 25%) and kinetin (0 mg/L, 1 mg/L, 3 mg/L). The results showed that there were no interaction between coconut water and kinetin concentration to the growth of initiation pamelo orange in vitro for all observed variables, except for the number of shoot, number of leaves and the number of roots. The treatment of coconut water concentration single effects the number of shoots, the height of the buds and the time of root growth. The effect kinetin concentrations treatment single only affects shoots growth time. Keywords: Coconut Water, Kinetin, Initiation, Pamelo, In Vitro
