Abstract

ABSTRACT Kawista (Limonia acidissima) is a plant that belongs to the citrus tribe (Rutaceae). Kawista is one of the natural ingredients that is beneficial for health. Kawista skin and pulp contain chemical compounds of alkaloids, flavonoids, saponins, tannins and terpenoids that function as antioxidants. The kawista plant is a plant that has begun to be rare with a decreasing population. The purpose of this study was to determine the effect of coconut water, kinetin and the interaction between coconut water and kinetin on the initiation of kawista fruit seeds in vitro. This research used a two-factor completely randomized design (CRD) method. The first factor was the provision of coconut water concentration (A), consisted of three levels, namely: A0 = 0%, A1 = 20%, A2 = 30%, A3 = 40%. The second factor was the provision of kinetin growth regulator (K) consisted of three levels, namely: K0 = 0 mg/L, K1 = 1 mg/L, K2 = 2 mg/L. The results showed thet the treatment of coconut water concentration affected the initiation of kawista seeds in vitro. This could be seen in the variable percentage of shoot growth 5-7 WAP, number of buds 5-7 weeks, shoot height and number of leaves 6-8 WAP. The best treatment was found in the treatment 0% coconut water concentration. Kinetin concentration treatment affected the initiation of kawista seeds in vitro. This could be seen in the variable number of buds 4 and 7 WAP after planting, number of leaves 7-8 WAP. The best treatment was found in the treatment 1 ppm kinetin concentration. There was no interaction between the coconut water concentration and Kinetin in act of the variables Keywords: Explant, LAFC, MS media, Plantlet.
