Abstract
ABSTRAK Dislipidemia merupakan gangguan metabolisme lipid yang dapat meningkatkan stres oksidatif dan menyebabkan kerusakan jaringan jantung. Daun kelor (Moringa oleifera) diketahui mengandung senyawa bioaktif, seperti flavonoid dan polifenol, yang berpotensi memiliki efek antioksidan dan kardiokuratif, sehingga perlu diteliti pengaruhnya terhadap kerusakan histopatologi jantung pada kondisi dislipidemia. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak daun kelor terhadap histopatologi jantung tikus putih jantan (Rattus norvegicus) model dislipidemia. Penelitian ini merupakan penelitian eksperimental murni dengan rancangan post-test only control group design yang dilakukan di Laboratorium Patologi Anatomi dan Laboratorium Farmakologi Fakultas Farmasi Universitas Sumatera Utara. Sampel penelitian terdiri atas 36 ekor tikus Wistar jantan yang dibagi menjadi 6 kelompok, yaitu kontrol normal, kontrol negatif, kontrol positif, serta tiga kelompok perlakuan ekstrak daun kelor dosis 250, 500, dan 750 mg/kgBB/hari. Hasil penelitian menunjukkan bahwa kelompok kontrol negatif mengalami kerusakan histopatologi paling nyata, terutama pada parameter nekrosis miokardium, sedangkan kelompok perlakuan P1, P2, dan P3 tidak menunjukkan peradangan interstisial, fibrosis interstisial, maupun nekrosis miokardium. Uji Kruskal-Wallis menunjukkan bahwa tidak terdapat perbedaan bermakna pada parameter peradangan interstisial dan fibrosis interstisial (p=0,416), namun terdapat perbedaan bermakna pada parameter nekrosis miokardium (p=0,004). Uji post hoc Dunn menunjukkan adanya perbedaan bermakna antara kelompok kontrol negatif dengan seluruh kelompok perlakuan. Berdasarkan hasil tersebut, dapat disimpulkan bahwa pemberian ekstrak daun kelor dosis 250, 500, dan 750 mg/kgBB/hari berpengaruh terhadap gambaran histopatologi jantung tikus putih jantan model dislipidemia, terutama pada parameter nekrosis miokardium. Kata kunci: ekstrak daun kelor; dislipidemia; histopatologi jantung; nekrosis miokardium; Rattus norvegicus iv ABSTRACT Dyslipidemia is a disorder of lipid metabolism that can increase oxidative stress and cause damage to cardiac tissue. Moringa oleifera leaves are known to contain bioactive compounds, such as flavonoids and polyphenols, which are considered to have antioxidant and cardioprotective effects; therefore, their effect on cardiac histopathological damage under dyslipidemic conditions needs to be investigated. This study aimed to determine the effect of Moringa oleifera leaf extract administration on the cardiac histopathology of male white rats (Rattus norvegicus) with dyslipidemia. This study was a true experimental study with a post-test only control group design, conducted at the Anatomical Pathology Laboratory and Pharmacology Laboratory, Faculty of Pharmacy, Universitas Sumatera Utara. The sample consisted of 36 male Wistar rats divided into 6 groups: normal control, negative control, positive control, and three treatment groups receiving Moringa oleifera leaf extract at doses of 250, 500, and 750 mg/kgBW/day. The results showed that the negative control group exhibited the most prominent histopathological damage, especially in the myocardial necrosis parameter, whereas the treatment groups P1, P2, and P3 showed no interstitial inflammation, interstitial fibrosis, or myocardial necrosis. The Kruskal-Wallis test showed no significant differences in interstitial inflammation and interstitial fibrosis (p=0.416), but a significant difference was found in myocardial necrosis (p=0.004). Dunn’s post hoc test showed significant differences between the negative control group and all treatment groups. Based on these findings, it can be concluded that administration of Moringa oleifera leaf extract at doses of 250, 500, and 750 mg/kgBW/day affected the cardiac histopathological features of male white rats with dyslipidemia, especially in the myocardial necrosis parameter.
